Conagen’s Grant Award Opens New Markets for Monoclonal Antibodies

Funding from the Bill & Melinda Gates Foundation for Conagen’s Conamax platform benefits developing countries and patients globally.

Bedford, Mass., March 29, 2022 (GLOBE NEWSWIRE) — The Bill & Melinda Gates Foundation has given a grant to Conagen to support further development of its Conamax(TM) platform for the production of accessible low-cost and high-quality monoclonal antibodies, benefitting developing countries and patients globally.

The high cost of monoclonal antibody (mAb) production makes blockbuster drugs expensive, limiting the markets in which these molecules can be applied and limiting access to large patient populations in developing and industrialized countries. The reason for high production costs is, in part, intrinsic to the use of mammalian cell expression systems for antibody manufacturing.

“The Conamax platform was originally conceived to address this global unmet need, so we are thrilled to have the support of the Bill & Melinda Gates Foundation to help develop affordable antibody therapeutics,” said Casey Lippmeier, Ph.D., vice president of innovation at Conagen.

With rapid cell growth, human-compatible glycan structures, and demonstrated world-scale fermentation bioprocesses, the Conamax platform holds several advantages over Chinese Hamster Ovary (CHO) cell lines and other mammalian expression systems, as well as other microbial platforms.

The foundation grant funds the bench-scale development of a proof-of-concept study of a continuous purification process, customized to inputs from Conamax and potentially other microbial host organisms. With large-scale advancements, Conagen envisions that this process will be capable of continuously purifying mAbs from material generated in bioreactors with volumes greater than 250,000 liters.

As a significant advantage, the process will not require expensive binding proteins or other expensive column purification steps. This process will enable economies of scale which are not accessible to CHO or other mammalian-derived cell systems while also providing rapid, high-throughput purification of large amounts of antibody.

“Incumbent antibody manufacturing and purification processes based on Chinese Hamster Ovary (CHO) cell lines have brought tremendous advancements to biopharmaceuticals,” said Lippmeier. “However, CHO cell systems are comparatively low volume, expensive, and do not enable low cost and efficient purification of large amounts of antibodies.”

According to McKinsey, in 2019, global sales revenue for all mAb drugs was nearly $163 billion, representing about 70% of the total sales for all biopharmaceutical products, approximately $230 billion. That is about a 50% growth in sales and proportion since 2013, when it was $75 billion.

Continued growth in sales of currently approved mAb products, along with more than 1,200 mAb product candidates currently in development — many for multiple indications — will continue to drive the overall sales of all biopharmaceutical products.

“We’re unlocking the way to make drugs more affordable and, while doing so, opening additional markets for biologics,” said Lippmeier.

Will Smith Apologizes to Chris Rock for Oscars Slap

Actor Will Smith apologized to comedian Chris Rock one day after slapping him across the face during Sunday’s Academy Awards ceremony following a joke Rock made about the actor’s wife, Jada Pinkett Smith.

Smith’s apology Monday came after the Academy of Motion Picture Arts and Sciences issued a formal statement condemning his behavior for slapping Rock and announced it was initiating an investigation.

“Violence in all of its forms is poisonous and destructive,” Smith said Monday in a statement his publicist posted on Instagram.

“I would like to publicly apologize to you, Chris. I was out of line and I was wrong. I am embarrassed and my actions were not indicative of the man I want to be,” Smith added.

The Academy of Motion Picture Arts and Sciences announced it was initiating an investigation into the incident. According to the Academy’s code of conduct, Smith could face consequences such as “suspension of membership or expulsion from membership.”

In recent years, violations of this code have led to the removal of members such as Harvey Weinstein, Bill Cosby and Roman Polanski, who were expelled over allegations of sexual misconduct.

Rock did not file a police report.

“LAPD investigative entities are aware of an incident between two individuals during the Academy Awards program. The incident involved one individual slapping another. The individual involved has declined to file a police report,” said the LAPD in an official statement to the press. “If the involved party desires a police report at a later date, LAPD will be available to complete an investigative report.

Ahead of presenting the award for best documentary, Rock directed a comment to Pinkett Smith saying: “Jada, I love you. ‘G.I. Jane 2,’ can’t wait to see it,” apparently drawing a comparison to actress Demi Moore’s shaved hairstyle in the 1997 movie G.I. Jane.

While Pinkett Smith was shown rolling her eyes following the comment, her husband walked over to the stage and slapped Rock across the face during the live broadcast, a moment that went viral shortly after.

After taking his seat again, Smith directed profanity at the comedian, demanding he not speak of his wife.

Pinkett Smith, 50, has previously been vocal about her struggles with hair loss due to the autoimmune disease alopecia. She publicly shared her diagnosis in 2018 during a segment of her Facebook talk show series Red Table Talk.

Initially, the Academy had only issued a statement via its Twitter account after the ceremony ended.

“The Academy does not condone violence of any form,” the tweet said.

Later Sunday evening, Smith was awarded an Oscar for Best Actor in a Leading Role for his performance as Richard Williams, the father of tennis champions Venus and Serena Williams, in the film King Richard.

During his acceptance speech, Smith appeared emotional and apologized to the Academy and his fellow nominees, but did not directly mention Rock.

“Love will make you do crazy things,” Smith said, addressing his behavior.

“Thank you. I hope the Academy invites me back.”

Kashmir Film Sharpens Political Divisions in India

The phenomenal box-office success of a new film set in 1990s Kashmir has sharpened political divisions in India and prompted a re-examination of a violent campaign against Hindus in the Muslim-majority region three decades ago.

“The Kashmir Files,” directed by Vivek Ranjan Agnihotri, depicts the flight of Kashmiri Hindus, known as “Pandits,” from the region in early 1990s. It is a fictional narrative about a college student who learns that his Kashmiri Hindu parents were killed by Islamist militants, not in an accident as he was told by his grandfather.

The film is being enthusiastically promoted by India’s ruling Bharatiya Janata Party (BJP), which pursues a Hindu nationalist agenda and has been accused of fomenting animus toward the nation’s 200 million Muslims as an electoral strategy.

Indian Prime Minister Narendra Modi personally met with the director and producer of the film immediately after its release and expressed his appreciation.

Celebrities and political leaders also have urged people to see the film. The Union minister for Women and Child Development, Smriti Irani, tweeted, “Watch … so that this history soaked in the blood of innocents may never repeat itself #TheKashmirFiles.”

A goods and services tax that boosts the price of movie tickets has been waived in most BJP-ruled states including some of India’s most populous. In the central state of Madhya Pradesh, police have been offered a day off work to watch the film. In the national capital territory of Delhi, however, Chief Minister Arvind Kejriwal rejected a demand from BJP legislators to declare the film tax-free, saying, “Well, put it on YouTube, it will be free.”

Sushil Chaudhary, the founder and chairman of the digital movie theater chain Picture Time DigiPlex, told VOA he was pleased that the controversial subject had been addressed in a film.

“And the storytelling was very different compared to other Indian films. The way the director handled the film — it was quite amazing, at the same time very sensitive. This film has huge impact and reminded me of the much-celebrated ‘Schindler’s List,’” a 1993 film about a German businessman who rescued more than 1,000 Jews from the Nazi Holocaust.

On social media, commenters have described the movie as “the most hard-hitting film” about Kashmir made to date.

BJP’s support of ‘The Kashmir Files’

The film also has detractors, many of them in the conflict-torn region of Jammu and Kashmir itself. While expressing appreciation for the movie’s dramatic qualities, these critics say it oversimplifies the complex history of the conflict, and that it offers a clichéd representation of Kashmiri Muslims.

“Hindu supremacists in India have weaponized the Kashmiri Pandit exodus” wrote Nitasha Kaul, a Kashmiri Pandit and novelist based in London.

“The movie dwells on Kashmiri Pandit suffering alone and makes ample use of Islamophobic tropes – all Muslims in the movie are violent, barbaric or lecherous,” she wrote.

She argued that the movie “feeds into cycles of hate and revenge. It collapses Kashmir’s history and politics into an Islamophobic morality tale that is palatable and profitable to Hindutva India.”

Ashok Swain, the head of the department of peace and conflict research at Sweden’s Uppsala University, told VOA he believes the film was made purely for political purposes by a Hindu right-wing filmmaker with support from the ruling authorities.

The purpose of the movie is not to tell the history or support the cause of displaced Kashmir pundits, Swain maintained, “but to make economic gains for the filmmaker and political gains for the ruling regime by selling Muslim hate in the country.”

Regional take

The movie also has been met with criticism by Muslim leaders of the Peoples Democratic Party, a Kashmir-based regional political bloc. PDP leader and former chief minister Mehbooba Mufti has accused the BJP of doing nothing for the Kashmiri Pandits who remained in Jammu and Kashmir.

Earlier, she said the move is “ill-intentioned” and will not contribute to healing old wounds.

Mufti also argued that while the filmmakers were mainly interested in profits, Modi and the BJP were supporting the film in order to instigate people along religious lines.

Kashmir resident Sameer Kaul told VOA that some of the gory incidents portrayed in the documentary-style film actually occurred, but that the movie falsely suggests the entire Kashmiri Muslim community played a part in the violence. In fact, he said, some Muslims opposed the violence and others were simply frightened.

Kaul said the impact of the movie will be to increase religious polarization and potential intercommunal discord. “Never before has the justification for institution of an unbiased judicial probe by central government seemed as convincing. Truth should hopefully pave the way for closure, reconciliation and desperate peace.”

A similar view has been expressed by one Kashmiri Pandit girl, Sagrika Kissu. “Not every Muslim is a terrorist/militant or a terrorist sympathizer,” she posted on social media. “We should be very sensitive when we paint all of them in one color. This movie sets in a very bitter emotion for Kashmiri Muslims as whole.”

Real-life impact

Meanwhile, the impact from the movie is being felt in real life. A hotel in Delhi recently refused to accommodate a Kashmiri man even after he provided appropriate identification and other credentials.

The hotel’s receptionist said the Delhi Police had told the hotel not to accept reservations from guests from Jammu and Kashmir. A video of the incident went viral, prompting Delhi Police to deny having issued any such order.

In an immediate reaction, the hotel chain Oyo Rooms removed the hotel from its platform.

Nevertheless, the film is doing blockbuster business despite a lack of promotion and marketing, appearing on 700 screens across India and grossing $3 million since its release on March 11. It is also being shown in the United States, Britain, Canada and Australia, taking in $1.38 million in its first week on international screens.

European Union funding boosts COVID-19 vaccination in Africa

Brazzaville/Copenhagen – The European Commission’s Directorate-General for European Civil Protection and Humanitarian Aid Operations (ECHO) is supporting the work of the World Health Organization (WHO) to boost COVID-19 vaccination campaigns and increase coverage in 15 African countries.

With a total of €16 million from ECHO, WHO is supporting operational and technical aspects of the vaccine rollout over 18 months in the following countries: Burundi; Cameroon; Central African Republic; Chad; Democratic Republic of the Congo; Guinea; Liberia; Madagascar; Mali; Mozambique; Niger; Nigeria; Somalia; South Sudan Republic, and Sudan. The grant is also helping to reinforce the capacity of health workers to plan, coordinate and deploy the vaccines as well as monitor and document the results of the rollout and adequately investigate and report any adverse events following immunization.

The funding is part of the European Union’s (EU) humanitarian initiative for COVID-19 vaccination in Africa which aims to ensure increased access including for the most vulnerable as well as those living in hard-to-reach, remote and conflict-affected areas. It is also one of the many contributions from the EU and its Member States to the COVID-19 response. The EU also supports the COVAX Facility, the vaccines part of the Access to COVID-19 Tools-Accelerator created to develop and deliver tools to fight the pandemic.

“Ever since the COVID-19 pandemic broke out, the EU, its Member States and European financial institutions have come together as Team Europe, contributing to the fight against the pandemic around the world, supporting in particular the African continent,” said Paraskevi Michou, Director General for Humanitarian Aid and Civil Protection in the European Commission. “In addition to being a leading donor to the COVAX Facility, the EU supports local manufacturing of medicines and vaccines, the strengthening of research, analysis and sequencing capacities, as well as the improvement of health systems at regional and national levels in Africa. The EU has also provided a total of €100 million in humanitarian assistance to specifically support the rollout of vaccination campaigns in Africa, to help ensure equitable access to vaccines for vulnerable people, including in conflict-affected or hard-to-access areas. Our strong cooperation with the World Health Organization is instrumental in implementing this programme successfully.”

Efforts are ongoing to scale up COVID-19 vaccine coverage in Africa, where only 15% of the population is fully vaccinated so far. Around 404 million of the more than 716 million doses the continent has received to date have been administered.

In a fresh drive to support countries’ scale up vaccination, WHO and partner organizations have deployed more than 60 experts on the ground to form part of country expert teams. These teams are working to strengthen coordination, logistical and financial planning, including microplanning, surveillance of adverse events following immunization as well as vaccine uptake and stock data management. WHO partners are also working with people in the communities to strengthen trust and confidence in vaccination.

“With African countries striving to expand the COVID-19 vaccination coverage, the support by the European Union injects a crucial momentum into the drive to scale up coverage on continent. Vaccination is the best protection against the adverse impact of the virus and will also prevent new variants from emerging and threatening not only Africa but the world.,” said Dr Matshidiso Moeti, WHO Regional Director for Africa.

The vaccination campaigns prioritize vulnerable and high-risk populations such as health workers, older people, those with co-morbidities, in particularly living in fragile, conflict-affected and humanitarian contexts, including in refugee camps.

“Solidarity is key to ending this pandemic and to building back better. These are not just words. These principles have been already exemplified by the generous support with vaccines and funding provided by the European Union to the global pandemic response. Together in Europe, in Africa and beyond, WHO and the EU are working with local partners to ensure COVID-19 vaccination reaches the arms of everyone and that lessons learned contribute to resilient health systems,” said Dr Hans Henri P. Kluge, WHO Regional Director for Europe.

As the continent battles the pandemic it is also crucial to step up the efforts to address other vaccine-preventable disease as well as bolster health systems to provide accessible, quality and affordable care.

Eritrea Observed World TB Day 2022

World TB Day was commemorated on 24 March 2022 in Asmara, the capital city of Eritrea. The theme of World TB Day for 2022 was “Invest to end TB. Save lives”. TB is one of the top 10 causes of death worldwide and millions of people continue to fall sick with TB each year.

Strong political commitment, a community-based health system, and high community engagement in Eritrea significantly contributed to the declining burden of TB. The country is implementing the global recommended TB control strategies and services that are provided in public, non-governmental, and non-profit private health facilities. Since the implementation of the strategies, many TB cases have been identified and treated successfully, and many more lives have been saved. In Eritrea, everyone has free access to TB treatment and management.

During the event, the Director of Communicable Disease Control in the Ministry of Health, Dr. Araia Berhane said that the Government of the State of Eritrea with its partners has scored great achievements in the prevention and management of susceptible and drug resistance tuberculosis though some challenges need still to be addressed. Furthermore, the Ministry of Health has put plans with evidence-based interventions to reduce TB prevalence to zero. Some of the interventions are:

• To initiate an outreach service to reach the hard-to-reach areas with specialized diagnosis to increase TB detection.

• To implement the management of susceptible TB and drug-resistant TB with 4 months of treatment and 6-9 months of treatment according to the updated WHO TB management guideline.

• To initiate MDR TB management at community through DOTS strategy

• To equip health facilities with GeneXpert machines including in remote areas.

At the end, Dr Araia appreciated WHO support and thanked MOH, MOE colleagues, teachers and students who made key TB presentations, and poems for increasing awareness in the community during the event.

On behalf of WHO Regional Director for Africa, Dr. Matshidiso Moeti and representing WHO Representative to Eritrea, Dr Martins Ovberedjo, Dr. Assefash Zehaie, ATM/Officer read RD’s message that to reach the target, TB incidence would have had to record an annual decline of 4% to 5% in 2020, increasing to 10% per year by 2025, and then to an average 17% annually in the following decade. She indicated that the world saw an increase in the number of global TB deaths for the first time in over a decade in 2021. Contributing factors to the deaths included reduced access to TB diagnosis and treatment, in the face of the COVID-19 pandemic.

She noted that with 36% of all TB deaths occurring in Africa, failure to invest in the TB response is set to take a formidable toll on African countries.

Therefore, the call on governments is to mobilize additional domestic financial support for TB control, including contributions to the Global Fund, which last month launched its US$18-billion Seventh Replenishment campaign in a bid to counter the catastrophic impact of COVID-19 on the fight against TB.

She urged all stakeholders to advocate for increased investment and to ensure that TB services are integrated into the primary health care response for maximum impact.

At the commemoration, higher officials from the Ministry of Health, Ministry of Information, Ministry of Education, representatives from the National Union of Eritrean Women, National Union of Eritrean Youth and Students, Health professionals, Students, teachers were present.

Prize winners of the contest who made presentations and read poems were awarded prizes.

What is impeding Africa’s TB fight?

Cape Town – Africa has made progress in the fight against tuberculosis (TB) in recent years, however several hurdles are curbing the efforts to end this preventable and curable disease and at the current pace, the global targets to eliminate the disease by 2030 look increasingly elusive. Dr Norbert Ndjeka, Chief Director of TB Control and Management in South Africa’s National Department of health, assesses the challenges and how to accelerate progress.

Why is progress against tuberculosis slowing down in Africa?

There are several factors, including the fact that TB screening and testing are not been appropriately done. Many countries in Africa still rely on smear microscopy which is less sensitive than the recommended WHO rapid diagnostic test. Limited ability to find people with TB explains the fact that TB transmission continues to increase. Universal access to drug susceptibility tests adds to the challenge. The gap between the estimated number of TB cases and the number of TB cases detected remains very wide. The introduction of new drugs has also been very slow due to numerous reasons, while the African region is also struggling to scale up of TB preventive therapy. Weak health systems, poverty and inadequate nutritional support in many countries also contribute to the high TB burden in the region. TB infection is influenced by social and economic development determinants such as undernutrition, diabetes, HIV infection, alcohol use disorders and smoking which must be addressed through a multisectoral action and accountability to ensure its timely implementation. Despite the toll of the disease, TB is not yet at the forefront of health priorities in many countries. In Africa, governments contribute only 22% of the resources required to deliver adequate tuberculosis services, while 44% of needs remain unfunded. There is an urgent need to intensify efforts to mobilize additional funds from domestic sources and international donors.

How has COVID-19 affected TB response in the continent?

COVID-19 has affected TB case finding and TB linkage to care. In many countries, human, financial and other resources have been reallocated from tackling TB to the COVID-19 response, limiting the availability of essential services. People affected by TB have struggled to seek care in the context of lockdowns. COVID-19 has also impacted the detection of drug-resistant TB, with the number of cases recorded in the AFRO region has decreased by 28% in 2020 compared to 2019.

There are also reports of inadequate patient retention in care, as COVID-19 has exacerbated weaknesses existing before the pandemic. Although there are negative effects, we can apply some of the lessons learned in fighting COVID-19 in TB control. These include the effective collaboration between the public and the private health sectors, the real-time reporting of new cases and effective dissemination of information. The consistent use of personal protective equipment’s (masks and respirators) is also something that we had wanted to do in health care facilities in high TB burden areas. We need to ensure this practice stays in health care settings in high-burden settings. Although to treat is to prevent, but prevention in the first place is better.

What can be done to reach the TB elimination targets?

We need to scale up TB preventive therapy. Improvement of infection control measures at all public and private health care facilities is required to prevent TB. Several people get TB by visiting health care facilities for other health conditions than TB. We need to make our health care facilities safe. We also need to implement our case finding strategies by using rapid molecular diagnostic tests as initial TB diagnostic tests. We must also adopt universal testing and treatment approach. By so doing, the gap between estimated TB numbers and TB detected cases can be reduced and it will be possible to curb TB transmission if patient-centred models of care are put in place. Treatment with new and shorter treatment regimens with higher efficacy and better safety should also be adopted by countries. Establishing strong partnerships with civil society and academia is necessary to enhance TB response. TB programmes in the region need to support operational research while protecting vulnerable people. Finally, we need to note that TB is also a social problem. Poverty reduction and improvement of general living conditions such as housing, employment will go a long way in decreasing TB transmission. A TB vaccine that protects against pulmonary tuberculosis is an imperative. That is why a multisectoral response is needed if we are to beat TB.
